Antiviral drug for use in HIV infection.
Ritonavir is an inhibitor of HIV-1 and HIV-2 aspartyl proteases for oral administration, an active peptidomimetic. Inhibition of HIV proteases prevents the polyprotein from breaking the gag-pol bond, resulting in the formation of an immature and incapable of infection virus. Ritonavir has a selective affinity for HIV protease and shows little activity against human aspartyl protease.
The antiviral activity of ritonavir in vitro was evaluated on lymphoblastic cell lines and lymphocytes to suppress viral replication by 50% (EC50) ranged from 0.0038 to 0.153 μmol, depending on the HIV-1 isolates used. In this case, the average value of EC50 was 0.022 µmol. In MT4 cells, the additive effect of ritonavir in combination with (ZDV) or didanosine (ddI) against HIV-1 has been shown.
Dosage
The drug should be taken orally during a meal.
In adults, the recommended dose is 600 mg (6 caps.) 2 times / day. A gradual increase in the dose of Norvir in the initial period of treatment may improve the tolerability of the drug.
