Release form of the drug Lerivon
Produced in the form of white rounded film-coated tablets. On one side of the tablet there is the Organon logo, on the back there is a transverse line for ease of dosing. The blister enclosed in a cardboard box contains 20 tablets. One tablet of Lerivon contains 30 mg of the active substance mianserin.
Pharmacological action of Lerivon tablets
Lerivon is classified as a tetracyclic antidepressant of the piperazino-azepine series.
By blocking alpha2-adrenergic receptors, the drug increases the concentration of the norepinephrine mediator in the synaptic cleft. Enhanced noradrenergic transmission of nerve impulses increases brain activity. This is how the anxiolytic activity of Lerivon is mediated.
Activation of alpha1-adrenergic and H1-histamine receptors causes a pronounced sedative effect of the drug.
Indications for the use of tablets Lerivon
Lerivon is effective against depressive disorders of various origins.
How to use Lerivon tablets
Lerivon tablets are taken orally, washed down with water and not chewed.
In adults, at the beginning of treatment, 30 mg of the drug is prescribed. Dose adjustment is made depending on the clinical effect. The maintenance dose should be less than the initial dose. The maximum allowable amount of Lerivon per day is 60–90 mg.
In elderly patients, at the initial stage of therapy, the same amount of Lerivon is prescribed. For the onset of a therapeutic effect, smaller doses are needed, which are titrated by the attending physician during the course of treatment.
In children and adolescents, Lerivon is not used due to the lack of objective data on the clinical experience of its use.
It is better to take the drug at bedtime because of its pronounced sedative effect.
The average duration of the course of treatment is 4-6 months. Antidepressant therapy is continued for several more months after the patient’s condition improves. Abrupt discontinuation of Lerivon is not recommended, as the risk of withdrawal syndrome increases.
Side effects of Lerivon tablets
Frequent undesirable manifestations of the action of Lerivon are disorders of the central nervous system: hypomania, convulsive readiness with hyperkinetic syndrome, neuroleptic malignant syndrome.
At the initial stage of taking the drug, a tendency to bradycardia and various arrhythmias is possible.
Pharmacokinetic features of the drug cause the appearance of increased activity of liver enzymes and jaundice in some cases.
In the blood picture, agranulocytosis and granulocytopenia are possible.
In some patients, arthralgia and an allergic rash occurred as adverse reactions to taking Lerivon.
